Uses
Indicated for metastatic non-small cell lung cancer (NSCLC) with EGFR exon 19 deletions or exon 21 (L858R) mutations. Also indicated as first-line treatment for locally advanced or metastatic pancreatic cancer in combination with gemcitabine.
Do not use if
No absolute contraindications listed. However, it should be used with extreme caution in patients with hepatic impairment, history of interstitial lung disease, or severe renal impairment.
Side effects
Common adverse reactions include rash, diarrhea, anorexia, fatigue, nausea, and dyspnea. Serious events may include interstitial lung disease, hepatotoxicity, renal failure, and gastrointestinal perforation.
Warnings
Monitor for signs of interstitial lung disease; discontinue if confirmed. Monitor liver function tests and renal function regularly. Risk of serious skin reactions including toxic epidermal necrolysis.
How it works
Erlotinib reversibly inhibits the kinase activity of EGFR, preventing autophosphorylation of tyrosine residues and blocking downstream signaling pathways involved in cell proliferation and survival. It is selective for EGFR-mutant tumors in NSCLC.
