Uses
Flibanserin is indicated for the treatment of acquired, generalized hypoactive sexual desire disorder (HSDD) in premenopausal women under 65 years of age, characterized by low sexual desire causing marked distress or interpersonal difficulty and not attributable to a co-existing condition, relationship problems, or medication effects.
Do not use if
Contraindicated with concomitant use of moderate or strong CYP3A4 inhibitors, in patients with hepatic impairment, and with known hypersensitivity to flibanserin or any component. Alcohol consumption around the time of dosing is contraindicated.
Side effects
Most common adverse reactions include dizziness, somnolence, nausea, fatigue, insomnia, and dry mouth. Severe hypotension and syncope may occur, especially when combined with alcohol or CYP3A4 inhibitors.
Warnings
Black box warning for hypotension and syncope when combined with alcohol or CYP3A4 inhibitors. Patients should be advised to avoid alcohol within two hours of dosing or when going to sleep. CNS depression may impair ability to drive or operate machinery.
How it works
Flibanserin is a serotonin 5-HT1A receptor agonist and 5-HT2A receptor antagonist with dopamine D4 receptor agonist activity. It modulates serotonin and dopamine neurotransmission in brain areas involved in sexual desire, though its precise mechanism in treating HSDD is not fully established.
