Uses

Thiamphenicol is a broad-spectrum antibiotic indicated for the treatment of susceptible bacterial infections including respiratory tract infections, urinary tract infections, and sexually transmitted infections such as gonorrhoea and chlamydia.

Do not use if

Contraindicated in patients with pre-existing haematological disorders, renal or hepatic impairment, and known hypersensitivity to thiamphenicol or chloramphenicol. Contraindicated in neonates and during pregnancy and breastfeeding.

Side effects

The most significant adverse effect is reversible bone marrow suppression causing anaemia, leukopenia, and thrombocytopenia. Gastrointestinal effects include nausea, vomiting, and diarrhoea. Unlike chloramphenicol, aplastic anaemia is rarely associated with thiamphenicol.

Warnings

Monitor blood counts during therapy due to risk of reversible bone marrow suppression. Avoid use in patients with severe renal impairment or those requiring high-dose or prolonged therapy without haematological monitoring.

How it works

Thiamphenicol is a semisynthetic broad-spectrum bacteriostatic antibiotic, a structural analogue of chloramphenicol. It inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit and blocking peptidyl transferase activity. It is active against a wide range of Gram-positive and Gram-negative organisms.
