Uses
Adjunctive treatment to levodopa/carbidopa in patients with Parkinson's disease experiencing motor fluctuations with 'off' episodes. It extends 'on' time without troublesome dyskinesia.
Do not use if
Contraindicated with concomitant use of other MAO inhibitors (including linezolid), opioids such as meperidine, and serotonergic agents. Also contraindicated in patients with severe hepatic impairment or albinism.
Side effects
Common adverse effects include dyskinesia, falls, nausea, insomnia, and hallucinations. Hypertension and, rarely, serotonin syndrome may occur. Excessive daytime sleepiness and sudden onset of sleep have been reported.
Warnings
Monitor blood pressure closely; may cause or worsen hypertension. Serotonin syndrome risk increases with concomitant serotonergic drugs. Advise patients about risk of somnolence and sudden sleep onset during activities such as driving.
How it works
Safinamide is a selective, reversible MAO-B inhibitor that reduces dopamine catabolism in the brain, thereby increasing dopaminergic tone. It also has sodium and calcium channel-blocking properties that may modulate glutamate release.
Storage
Store at 25°C (77°F); excursions permitted between 15°C and 30°C. Keep in the original container, protected from light and moisture.
