Uses
Indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes mellitus. Not recommended for type 1 diabetes or diabetic ketoacidosis.
Do not use if
Contraindicated in patients with NYHA Class III or IV heart failure and in those with a history of serious hypersensitivity to alogliptin or pioglitazone.
Side effects
Common adverse effects include upper respiratory tract infection, nasopharyngitis, and peripheral edema. Serious risks include congestive heart failure, pancreatitis, hypersensitivity reactions, and hepatic effects.
Warnings
Thiazolidinediones including pioglitazone can cause or worsen congestive heart failure; monitor closely after initiation. Risk of pancreatitis; discontinue if suspected. Risk of bladder cancer with prolonged use of pioglitazone.
How it works
Alogliptin inhibits DPP-4, increasing incretin hormones (GLP-1 and GIP) that stimulate glucose-dependent insulin secretion. Pioglitazone is a PPAR-gamma agonist that improves insulin sensitivity in peripheral tissues and the liver.
Storage
Store at 25°C (77°F); excursions permitted to 15–30°C (59–86°F). Keep container tightly closed and protect from moisture and humidity.
