Uses

Ixazomib is indicated in combination with lenalidomide and dexamethasone for the treatment of adult patients with multiple myeloma who have received at least one prior therapy. It is not recommended for maintenance therapy or newly diagnosed multiple myeloma outside of controlled clinical trials.

Do not use if

No absolute contraindications are listed in the product labeling. Use with caution in patients with hepatic or renal impairment and adjust dose accordingly.

Side effects

Common adverse reactions include thrombocytopenia, diarrhea, constipation, nausea, vomiting, peripheral neuropathy, peripheral edema, and skin rashes. Serious cutaneous reactions such as Stevens-Johnson syndrome have been reported.

Warnings

Monitor platelet counts at least monthly; dose adjustments required for thrombocytopenia. Adjust dosing for severe gastrointestinal toxicities. Monitor for peripheral neuropathy and edema. Embryo-fetal toxicity risk; effective contraception required for both sexes during and after treatment.

How it works

Ixazomib is an oral, reversible proteasome inhibitor that preferentially binds and inhibits the chymotrypsin-like activity of the beta-5 subunit of the 20S proteasome. This inhibition induces apoptosis in multiple myeloma cell lines and demonstrates cytotoxicity against relapsed/refractory myeloma cells.

Storage

Store at room temperature, not above 30°C (86°F). Do not freeze. Keep capsules in the original packaging until immediately before use.
