Uses
Pimecrolimus 1% cream is indicated as second-line therapy for mild to moderate atopic dermatitis in non-immunocompromised adults and children 2 years of age and older who have not responded adequately to other topical treatments.
Do not use if
Contraindicated in patients with hypersensitivity to pimecrolimus or any component of the cream. Not for use in immunocompromised patients. Avoid use in children under 2 years of age.
Side effects
The most common adverse reactions include application site burning or stinging, headache, nasopharyngitis, and upper respiratory infections. These local reactions tend to decrease over time with continued use.
Warnings
There is a boxed warning regarding the long-term safety of topical calcineurin inhibitors. Although a causal link has not been established, rare cases of malignancy have been reported. Use the minimum amount necessary and avoid continuous long-term use.
How it works
Pimecrolimus is a calcineurin inhibitor that binds to macrophilin-12 (FKBP-12), inhibiting calcineurin phosphatase activity. This prevents the dephosphorylation and nuclear translocation of NFAT, thereby reducing the production of inflammatory cytokines in T-cells.
Storage
Store at 25°C (77°F); excursions permitted between 15–30°C. Do not freeze.
