Uses
Poractant alfa is a porcine-derived natural lung surfactant indicated for rescue treatment of Respiratory Distress Syndrome (RDS) in premature infants. It reduces mortality and the incidence of pneumothorax associated with RDS.
Do not use if
No absolute contraindications have been identified.
Side effects
Common adverse reactions include bradycardia, hypotension, endotracheal tube blockage, and oxygen desaturation occurring transiently during administration. These are generally managed by temporarily interrupting the infusion.
Warnings
Administer only by trained personnel with immediate access to ventilatory support. Continuously monitor heart rate and oxygen saturation during administration. Rapidly adjust ventilator settings as lung compliance may change acutely after dosing.
How it works
Poractant alfa is a modified natural surfactant extracted from porcine lungs containing phospholipids and hydrophobic proteins (SP-B and SP-C). It replaces deficient endogenous surfactant in premature lungs, reduces alveolar surface tension during expiration, and prevents alveolar collapse, thereby improving lung compliance and gas exchange.
Storage
Refrigerate at 2–8°C. Protect from light. Unopened vials may be stored at room temperature (below 25°C) for up to 24 hours. Do not re-refrigerate after warming.
