Uses

Sulindac is a non-steroidal anti-inflammatory drug indicated for the treatment of osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, acute painful shoulder (subacromial bursitis or supraspinatus tendinitis), and acute gouty arthritis. It may be used for acute or long-term management.

Do not use if

Contraindicated in patients with hypersensitivity to sulindac or its excipients, and in those who have experienced asthma, urticaria, or allergic reactions to aspirin or other NSAIDs. Should not be used for perioperative pain in the setting of coronary artery bypass graft surgery.

Side effects

Common adverse effects include gastrointestinal disturbances such as nausea, dyspepsia, and abdominal pain. Serious effects include GI bleeding, peptic ulceration, cardiovascular thrombotic events, renal impairment, and hepatotoxicity. Skin reactions and hypersensitivity may occur.

Warnings

NSAIDs including sulindac increase risk of serious cardiovascular thrombotic events, myocardial infarction, and stroke, which may be fatal. Risk of serious GI adverse events including bleeding, ulceration, and perforation is increased. Use the lowest effective dose for the shortest duration.

How it works

Sulindac is a prodrug converted to an active sulfide metabolite that inhibits cyclooxygenase enzymes, reducing prostaglandin synthesis. This mechanism underlies its anti-inflammatory, analgesic, and antipyretic effects. Oral bioavailability is good with enterohepatic recirculation.

Storage

Store at 20°C to 25°C (68°F to 77°F). Dispense in a tight, light-resistant container.
