Uses
Emicizumab is a bispecific antibody indicated for routine prophylaxis to prevent or reduce bleeding episodes in adult and pediatric patients (including newborns) with hemophilia A (congenital factor VIII deficiency) with or without factor VIII inhibitors.
Do not use if
No formal contraindications. Caution is required when using with activated prothrombin complex concentrate due to significant risk of thrombotic microangiopathy and thromboembolism.
Side effects
Most common adverse reactions (≥10%) include injection site reactions, headache, and joint pain (arthralgia). Serious adverse events include thrombotic microangiopathy and thromboembolic events, particularly when used with activated prothrombin complex concentrate (aPCC).
Warnings
Risk of life-threatening thrombotic microangiopathy and thromboembolism when combined with aPCC at cumulative doses exceeding 100 U/kg/24 hours. Monitor closely if aPCC is required. Not a substitute for factor VIII in breakthrough bleeding management.
How it works
Emicizumab bridges activated factor IXa and factor X, mimicking the cofactor function of activated factor VIII. This restores effective hemostasis in patients with hemophilia A regardless of the presence of inhibitors. It is administered subcutaneously and exhibits dose-proportional pharmacokinetics.
Storage
Store in a refrigerator at 2°C to 8°C (36°F to 46°F) in the original carton to protect from light. Do not freeze or shake.
