Uses

Leuprolide acetate is indicated for management of endometriosis, uterine fibroids (leiomyomata), palliative treatment of advanced prostate cancer, and treatment of central precocious puberty in children.

Do not use if

Contraindicated in hypersensitivity to GnRH analogs or leuprolide acetate, undiagnosed abnormal uterine bleeding, and pregnancy. Not for use in women who are breastfeeding.

Side effects

Common side effects include hot flashes, decreased libido, vaginal dryness, bone mineral density loss with long-term use, and injection-site reactions. Initial flare of disease symptoms may occur at treatment start.

Warnings

Risk of bone mineral density loss; limit duration of therapy and consider add-back therapy when using for endometriosis. Initial flare of symptoms may transiently worsen prostate cancer. Monitor for psychiatric symptoms and convulsions.

How it works

Leuprolide is a GnRH agonist. Continuous administration causes downregulation of GnRH receptors and desensitization of the pituitary, leading to suppression of gonadotropin (LH and FSH) release and consequent reduction in sex hormone production (testosterone and estradiol).
