Uses

Pancuronium is a non-depolarizing neuromuscular blocking agent used as an adjunct to general anesthesia to facilitate endotracheal intubation and to provide skeletal muscle relaxation during surgery or mechanical ventilation.

Do not use if

Contraindicated in patients with known hypersensitivity to pancuronium or bromide. Use with caution in patients with cardiac disease due to vagolytic and sympathomimetic effects causing tachycardia.

Side effects

May cause tachycardia, hypertension, and increased salivation due to its vagolytic properties. Prolonged neuromuscular blockade may occur, especially in patients with renal or hepatic impairment. Hypersensitivity reactions are rare.

Warnings

Must be administered by trained anesthesia personnel only. Resuscitation equipment must be immediately available. Dose adjustments required in renal and hepatic impairment as the drug is primarily eliminated via the kidneys. Avoid in patients with myasthenia gravis.

How it works

Pancuronium is a bisquaternary aminosteroid that competitively antagonizes acetylcholine at the nicotinic receptors of the neuromuscular junction, producing flaccid paralysis of skeletal muscles. It has vagolytic activity leading to tachycardia and possesses some sympathomimetic properties.

Storage

Store under refrigeration between 2°C and 8°C. May be stored at room temperature for up to 6 months according to some formulations. Protect from light and freezing.
