Uses

Ruxolitinib is indicated for treatment of intermediate or high-risk myelofibrosis (primary or post-PV/ET), polycythemia vera in adults with inadequate response or intolerance to hydroxyurea, and steroid-refractory or steroid-dependent acute and chronic graft-versus-host disease.

Do not use if

No absolute contraindications are listed in the prescribing information. However, use is not recommended in patients with active serious infections until the infection is controlled.

Side effects

Common adverse reactions include thrombocytopenia, anemia, neutropenia, bruising, dizziness, and headache. Serious risks include opportunistic infections, non-melanoma skin cancers, and rebound of disease symptoms upon discontinuation.

Warnings

Risk of serious infections including tuberculosis and opportunistic infections. Monitor blood counts regularly; manage cytopenias with dose reduction or interruption. Avoid abrupt discontinuation to prevent rebound symptoms. Risk of secondary malignancies.

How it works

Ruxolitinib is a potent and selective JAK1/JAK2 inhibitor. Inhibition of JAK1/JAK2 disrupts cytokine signaling critical for hematopoiesis and immune function via the JAK-STAT pathway, reducing pro-inflammatory cytokine activity and aberrant cell proliferation.

Store at room temperature 20–25°C (68–77°F). Dispense in original container.
