Uses

Crizotinib is a kinase inhibitor indicated for adult patients with metastatic non-small cell lung cancer (NSCLC) whose tumors are ALK-positive or ROS1-positive. It is also indicated for pediatric patients aged 1 year and older and young adults with relapsed or refractory systemic ALK-positive anaplastic large cell lymphoma (ALCL).

Do not use if

No formal contraindications listed in the FDA label. However, use with caution in patients with pre-existing hepatic impairment, cardiac conduction abnormalities, or at risk of QT prolongation.

Side effects

Clinically significant adverse reactions include hepatotoxicity, interstitial lung disease/pneumonitis, QT interval prolongation, bradycardia, and severe visual loss. Common side effects include nausea, vomiting, diarrhea, edema, and fatigue.

Warnings

Fatal hepatotoxicity has been reported; monitor liver function tests periodically. Permanently discontinue in patients with interstitial lung disease/pneumonitis. Monitor ECG for QT prolongation and heart rate for bradycardia. Promptly evaluate any new visual symptoms as severe visual loss has occurred.

How it works

Crizotinib is an inhibitor of receptor tyrosine kinases including ALK, c-Met (HGFR), ROS1, and RON. Chromosomal translocations involving the ALK gene lead to expression of oncogenic fusion proteins that activate these kinases. Crizotinib inhibits these kinases, blocking tumor cell proliferation and survival signaling.

Storage

Store at room temperature between 20°C and 25°C. Keep in original container with tight closure. Keep away from children.
