Uses
Rimantadine hydrochloride is indicated for prophylaxis and treatment of illness caused by influenza A virus strains in adults aged 17 years and older. It is also indicated for prophylaxis against influenza A in children aged 1 to 16 years.
Do not use if
Contraindicated in patients with known hypersensitivity to rimantadine, amantadine, or other adamantane-class drugs.
Side effects
The most commonly reported adverse effects involve the gastrointestinal system (nausea, vomiting, abdominal pain) and the nervous system (insomnia, dizziness, nervousness, impaired concentration). These effects are generally mild to moderate.
Warnings
Use with caution in patients with epilepsy or other seizure disorders, as seizures have been reported. Dose reduction may be required in patients with severe hepatic or renal impairment. Resistance to rimantadine among influenza A strains may limit clinical efficacy.
How it works
Rimantadine inhibits influenza A viral replication by blocking the M2 ion channel, which prevents uncoating of the viral RNA within the host cell. It acts early in the viral replicative cycle and has no significant activity against influenza B.
