Uses

Alkylating antineoplastic used for newly diagnosed glioblastoma with radiotherapy and maintenance therapy. Also used for anaplastic astrocytoma in appropriate adult patients.

Do not use if

Contraindicated in patients with serious hypersensitivity to temozolomide, dacarbazine, or formulation components.

Side effects

Common effects include nausea, vomiting, fatigue, alopecia, and myelosuppression. Serious bone marrow suppression and liver toxicity may occur.

Warnings

Monitor blood counts and liver function regularly. Risk of severe myelosuppression, hepatotoxicity, opportunistic infections, and secondary malignancies.

How it works

Temozolomide is converted nonenzymatically to MTIC, which alkylates DNA and damages tumor cells, leading to cell death.
