Uses

Axitinib is a selective VEGFR-1, VEGFR-2, and VEGFR-3 inhibitor indicated for advanced renal cell carcinoma (RCC): as first-line therapy in combination with avelumab or pembrolizumab, or as a single agent following failure of one prior systemic therapy.

Do not use if

No formal contraindications listed in the FDA label. However, use with caution in patients with uncontrolled hypertension, recent arterial thromboembolic events, or significant hepatic impairment.

Side effects

Common adverse effects include hypertension, fatigue, diarrhea, decreased appetite, nausea, dysphonia, hand-foot syndrome, and mucositis. Serious risks include arterial and venous thromboembolic events, hemorrhage, cardiac failure, and reversible posterior leukoencephalopathy syndrome (RPLS).

Warnings

Monitor blood pressure and treat hypertension before and during therapy. Evaluate for thromboembolic events, hemorrhage, and cardiac dysfunction. Temporarily discontinue if signs of RPLS occur. Thyroid function should be monitored throughout treatment.

How it works

Axitinib potently and selectively inhibits VEGFR-1, VEGFR-2, and VEGFR-3, blocking tumor angiogenesis and proliferation by disrupting VEGF-mediated endothelial cell growth and survival signaling.

Storage

Store at room temperature between 20°C and 25°C (68°F to 77°F). Keep in the original container.
