Uses
Pazopanib is a multi-targeted tyrosine kinase inhibitor indicated for adults with advanced renal cell carcinoma (RCC) and advanced soft tissue sarcoma (STS) who have previously received chemotherapy. It is not effective for adipocytic soft tissue sarcoma or gastrointestinal stromal tumors.
Do not use if
No formal contraindications are listed in the FDA label. However, caution is required in patients with severe hepatic impairment, pre-existing QT prolongation, and significant cardiovascular disease.
Side effects
Common adverse effects include hepatotoxicity, hypertension, diarrhea, nausea, fatigue, hair color changes, and nausea. Serious risks include QT prolongation, cardiac dysfunction, hemorrhagic events, arterial thromboembolic events, and venous thromboembolism.
Warnings
Carries a boxed warning for severe and fatal hepatotoxicity; liver function must be monitored before and during treatment. Additional warnings include QT prolongation, cardiac dysfunction, hemorrhage, arterial and venous thromboembolism, and hypertension.
How it works
Pazopanib inhibits multiple receptor tyrosine kinases including VEGFR-1, VEGFR-2, VEGFR-3, PDGFR-α and -β, FGFR-1 and -3, Kit, Itk, and Lck. By blocking these receptors, it inhibits tumor angiogenesis and proliferation.
Storage
Store at room temperature between 20°C and 25°C (68°F to 77°F). Keep in the original container.
